The mechanism of action of the tadalafil citrate salt is due to a compound known as sildenafil which acts as a competitive antagonist for the 5-HT-2A receptor. The inhibitory effects of the compound on cyclic GMP in human cortical and striatal neurons are mediated mainly by the activation of the 5-HT-2A receptor. It has been shown that sildenafil, in addition to its main inhibitor, does not have any direct or indirect inhibitory effects on cGMP or phosphodiesterase (PDE)-5. In a series of experiments involving the reduction of cGMP by tadalafil in vitro, inhibition of cGMP by sildenafil was shown to be more potent than the inhibition of phosphodiesterase-5. The aim of the present study was to test the hypothesis that the action of the compound on the cGMP-PDE-5 receptor is due to its pharmacological effect or a direct inhibitory effect. The results indicate that the sildenafil metabolite, dapoxetine, has a direct inhibitory effect on the cGMP-PDE-5 receptor in vitro. This compound does not affect the phosphodiesterase-5 or cGMP-PDE-5 receptor in vivo. In a similar series of experiments, sildenafil and dapoxetine were tested in vitro against the cGMP-PDE-5 receptor in striatum and, after a 6-hour washout period, the cGMP-PDE-5 receptor was blocked by the compound at various doses (0, 1, 2, and 4 mg of tadalafil in a dose-dependent manner) and, following a 6-hour washout period, the cGMP-PDE-5 receptor was blocked by the compound at various doses (0, 1, 2, and 4 mg of tadalafil in a dose-dependent manner). The inhibition of the cGMP-PDE-5 receptor by the compound at various doses was also blocked by the compound at various doses (0, 1, 2, and 4 mg of tadalafil in a dose-dependent manner).
